Introduction
Affinity chromatography involves the property of bio recognition for the separation of proteins
based on the reversible interaction between the protein and the specific ligand bound to a
chromatography matrix. It enables us to purify bio molecule on the basis of its biological function
and its chemical structure. The goal of the experiment was to gain hands on experience in protein
purification by using AKTA FPLC system. The IgG antibody contained in the mouse antiserum was
purified by protein G affinity chromatography using a HiTrap protein G HP column. The
chromatogram was then obtained and analyzed to ensure the separation of IgG antibody. Such
purification of the protein before sample preparation is usually done to avoid any undesired
interactions.
Methods
The IgG antibody was separated by using AKTA FPLC by setting the method as: Column
HiTrap protein G HP 1 mL; Column pressure limit 0.300 MPa; Flow rate 1 mL/minute; Sample
injection 0.200 mL; Loading buffer, A 0.05 M sodium phosphate, 0.15M NaCl, pH 7.0; Elution
buffer, B 0.1 M glycine HCl, pH 2.7; Neutralization buffer, C 1 M Tris HCl, pH 9.0. Different
column volume for individual step was then set as 1 CV 0% B wash and equilibration step; 1

of the arts began to take hold. Romanticism, as the era was known, shifted the focus from rational

Classicalism. William Wordsworth, one of the numerous promulgators of this era, had set the stage,
writing over the complexity and nature of the world around him. One the most notable poems of

As a matter of personal opinion, this poem seems peculiar with its prophetic undertones. To
explain, a social divide like the one that is depicted by Wordsworth has been taking place in the
United Kingdom. Brexit , a referendum to leave the European Union that passed during the
summer of 2016, mimics the nationalistic fervor that is exhibited in the poem, especially in
phrasing such as ...raise us up, return to us again... (Meyer 703). In addition, one can infer that
since older generations of voters swayed the decision, they too have a longing for a revived golden
age, much like the plea that Wordsworth has towards the likes of John Milton, who was before
Wordsworth time. Of course, this notable poem could not only be directed to the United
Kingdom, but across the world as well. Across the many systems of governments, an ideological
shift towards the right on the political spectrum has offered growth in terms of their own nation
being the first of concern, and the rest of the world second. Such a shift tries to revive the golden
age that Wordsworth has longed desired. In essence, the poem exhibits a striking resemblance to the

In The Author to Her Book, Bradstreet is inundated in indecision and internal struggles over the
virtues and shortfalls of her abilities and the book that she produced. As human beings we associate
and sympathize with each other through similar experiences. It is difficult to sympathize with
someone when you don t know where they are coming from and don t know what they are dealing
with. Similar experiences and common bonds are what allow us to extend our sincere appreciation
and understanding for another humanbeing s situation. In this poem an elaborate struggle between
pride and shamemanifests itself through an extended metaphor in which she equates her book to
her own child.

She constantly speaks directly to her work as if it were her own child. Then in line 23 Bradstreet
calls herself the mother of this work.
Lines two through five move past depicting her as the mother and express how she feels
embarrassed that her works were published before she was ready to share them with the world
and without her consent. She says that the child had been by her side until snatched from thence
by friends, less wise than true (line 3). Basically she is saying a trusted person snatched her
work from her without permission to take them to England to be printed. Had it not been for her
brother in law taking her work back to England and getting them printed they may have never
been known. The intimacy and feeling she shares with her work is like that of a mother and
child and that bond was infringed upon when her work was exposed to public view (line 4). The
intrusion of her brother in law getting her work printed is the cause of feeling that follow.
Ironically the next thing she talks is the shame she has been thrust upon her by not being able to
perfect the work before it was published. This is illustrated in line five where she writes, Made thee
in rags, as to say her work is like a child dressed in rags.
